کاربرد نانو سلولز و هیدروکسی پروپیل متیل سلولز در پوشش دهی قرص کپستابین با هدف دارو رسانی به روده بزرگ

چکیده
بیان مسئله و اهداف: استفاده از پلیمرهای طبیعی برای انتقال دارو به دلیل سهولت دسترسی، ارزانی نسبی، پتانسیل تخریب و زیست-سازگاری یک حوزه فعال پژوهشی است. در این مطالعه قابلیت نانوکریستال سلولز(CNC)، نانو الیاف سلولز(CNF) و هیدروکسی‌پروپیل متیل سلولز(HPMC) به صورت پوشش قرص در سیستم دارو رسانی به روده بزرگ بررسی شده است.
مواد و روش‌ها: ابتدا قرص‌هایی از ترکیب کپستابین با شعاع و ضخامت 5/1 میلی‌متر ساخته شد. سپس این قرص‌ها به صورت دولایه توسط نانوکریستال سلولز و نانو الیاف سلولز با ضخامت 200 – 50 میکرومتر به عنوان لایه زیری و هیدروکسی پروپیل متیل سلولز با ضخامت 100 میکرومتر به عنوان لایه رویی پوشش دهی شدند. سپس انحلال قرص‌ها با شبیه سازی شرایط اسیدی معده و شبیه سازی شرایط pH خنثی و نزدیک به قلیایی روده کوچک بررسی شد. قرص هایی که در این شرایط سالم مانده بودند در شرایط میکروبی مشابه روده بزرگ و بدون میکروب از لحاظ حلالیت آزمایش شدند. همچنین مقاومت به کشش و شکنندگی قرص‌های ساخته شده به عنوان شاخص‌های مهم کیفی اندازه‌گیری شدند.
نتایج: نتایج آزمون انحلال نشان داد که قرص‌های کپستابین دارای پوشش نانوکریستال سلولز و نانو الیاف سلولز با ضخامت200 میکرومتر در لایه اول در شرایط میکروبی در زمان حدود یک ساعت دستخوش فروپاشی شدند اما در شرایط بدون میکروب تمامی نمونه ها در این ضخامت لایه پوشش سالم ماندند و فروپاشی انجام نشد. این نشان می‌دهد که شرایط میکروبی نقش موثری در تخریب و فروپاشی لایه پوشش نانوکریستال سلولز و نانو الیاف سلولز ایفا می‌کند. همچنین پوشش‌های مبتنی بر نانوسلولز - هم CNC و هم CNF به طور قابل توجهی استحکام کششی قرص‌های دارویی را افزایش دادند و این بهبود با ضخامت پوشش نسبت مستقیم نشان داد. مقدار شکنندگی قرص‌ها تابع لایه پوشش رویی هیدروکسی پروپیل متیل سلولز بوده و همگی در یک دامنه مشابه اندازه‌گیری شدند.
نتیجه‌گیری: به طور کلی نتایج این تحقیق نشان می‌دهد که پوشش‌دهی قرص‌های کپستابین با نانوسلولز هم به صورت نانو کریستال و هم نانوالیاف با ضخامت 200 میکرومتر به عنوان لایه اول و هیدروکسی پروپیل متیل سلولز با ضخامت 100 میکرومتر به عنوان لایه رویی قابلیت حفاظت از ترکیب کپستابین از شرایط تخریبی قسمت فوقانی دستگاه گوارش را دارد. این روش جدید می‌تواند به عنوان یک سیستم موثر دارو رسانی به روده بزرگ عمل کند. علاوه بر این شبکه نانوسلولز به طور فزاینده‌ای موجب بهبود یکپارچگی مکانیکی قرص‌های پوشش‌دار شد.
